- The studies reported in this investigation involved the isolation and biochemical characterization of galactosyltransferase (GalTase) from normal and physiologically altered (lactating) rat mammary gland plasma membrane isolates. Plasma membrane fractions, as verified by biochemical analyses, were isolated and characterized via Western-blot analysis and enzyme assays. Western blot analysis validated the presence of GalTase in plasma membrane fractions of both types of tissue isolates. However, there were marked changes in the kinetic activities of plasma membrane-associated GalTase in lactating tissue. In comparison studies of normal and lactating mammary gland plasma membrane-associated GalTase, the lactating tissue showed significantly decreased activity in reactions with tritiated-UDP-galactose when compared to the activity in normal tissue. The results generated from this study allowed for the proposal of a plausible cell-cell contact model.
