- Absorption and circular dichroism (CD) studies were performed on deoxy-HbA in the presence of various antisickling agents. In the presence of methylurea and butylurea, the absorption and CD studies on deoxy-HbA showed small changes at the absorption and CD maxima. However, the extent of these changes caused by the alky-urea could not be determined since experiments using the same HbA batch solution with the same amount of sodium dithionite added to it in different cells caused small differences at the absorption maxima. When sodium cyanate was added to deoxy-HbA at a concentration ratio ((NaCNoj/[deoxy-HbA]) of 1000 or greater, the deoxy-HbA peak at 43Onm shifted to 4l9nm and two peaks at 540nm and 57 Onm appeared in the absorption spectrum. In the CD spectrum, peaks appear at 42Onm in the Soret region and at 53Onm and 57Onm in the visible region when the NaCNO:deoxy-HbA concentration ratio equals 5000.
